eakon = air condition
kaketa = turned on
heya = room
ichiban = best/most
sugoshi = form of "sugosu" which means "to spend" as in "spend time in room"
yasui = peaceful/easy

The sentence in itself means "It's easiest(/coolest) to be in the room with the airconditioner"

You can also put it as:
It is most comfortable to be in a room with the air conditioner turned on.
